摘要 目的:通过建立小鼠味觉转化模型,研究多巴胺转运体(dopamine transporter, DAT)对这一行为的作用。方法:使用20只DATIRES CRE +/-的杂合小鼠和20只DATIREScre-/-的对照小鼠进行一系列味觉相关的行为学测试,通过Western Blot检测小鼠体内DAT蛋白在不同脑区的表达情况。结果:DATIREScre杂合组和对照组相比,其先天性味觉选择以及营养感受与分辨没有异常,但是在味觉感受转化时,尤其是在厌恶性味觉向偏好性味觉的转化中出现异常。结论:消化后营养感受需要多巴胺系统参与,多巴胺转运体表达降低会干扰这一重要过程。这可能是临床上注意力缺失过动症儿童挑食症状的潜在机制。

Abstract: Object: To analyze the influence of DAT (dopamine transporter) hypoexpression in the taste-to-nutrition suitability. Method: 20 DATIREScre heterozygous and littermates wild type controls were tested with series of taste behavior assays, also western blot was applied to measure the DAT expression in different brain regions. Results: Compared with the littermate control, heterozygous mice showed normal taste and nutrition sensation and preference; however, they expressed abnormalities when the taste and post-ingestive nutrition were mismatched. Conclusion: Dopamine information is necessary for coding nutrition valances. DAT hypoexpression interferes the nutritional sensing. This may be the potential mechanism of anorexia in the attention deficit hyperactivity disorder.
